ABSTRACT

ABSTRACT BACKGROUND: Serotonin (5-HT) is present in the epithelial enterochromaffin cells (EC), mast cells of the lamina propria and enteric neurons. The 5-HT is involved in regulating motility, secretion, gut sensation, immune system and inflammation. OBJECTIVE: Evaluate the effects of diabetes and quercetin supplementation on serotoninergic cells and its cell loss by apoptosis in jejunal mucosa of streptozotocin-induced diabetic rats (STZ-rats). METHODS: Twenty-four male Wistar rats were divided into four groups: normoglycemic (C), normoglycemic supplemented with 40 mg/day quercetin (Q), diabetic (D) and diabetic supplemented with 40 mg/day quercetin (DQ). After 120 days, the jejunum was collected and fixated in Zamboni's solution for 18 h. After obtaining cryosections, immunohistochemistry was performed to label 5-HT and caspase-3. Quantification of 5-HT and caspase-3 immunoreactive (IR) cells in the lamina propria, villi and crypts were performed. RESULTS: The diabetic condition displayed an increase of the number of 5-HT-IR cells in villi and crypts, while decreased number of these cells was observed in lamina propria in the jejunum of STZ-rats. In the diabetic animals, an increased density of apoptotic cells in epithelial villi and crypts of the jejunum was observed, whereas a decreased number of caspase-3-IR cells was observed in lamina propria. Possibly, quercetin supplementation slightly suppressed the apoptosis phenomena in the epithelial villi and crypts of the STZ-rats, however the opposite effect was observed on the 5-HT-IR cells of the lamina propria. Quercetin supplementation on healthy animals promoted few changes of serotoninergic function and apoptotic stimuli. CONCLUSION: These results suggest that quercetin supplementation mostly improved the serotonergic function affected by diabetes maybe due to antioxidant and anti-inflammatory properties of quercetin.


RESUMO CONTEXTO: A serotonina (5-HT) está presente nas células epiteliais enterocromafins (CE), nos mastócitos da lâmina própria e nos neurônios entéricos. A 5-HT está envolvida na regulação da motilidade, secreção, nocepção intestinal, sistema imunológico e inflamação. Objetivo: Avaliar os efeitos do diabetes e da suplementação de quercetina sobre a função serotoninérgica e a perda celular por apoptose na mucosa jejunal de ratos diabéticos induzidos por estreptozotocina (ratos STZ). MÉTODOS: Vinte e quatro ratos Wistar machos foram divididos em quatro grupos: normoglicêmico (C), normoglicêmico suplementado com quercetina 40 mg/dia (Q), diabético (D) e diabético suplementado com quercetina 40 mg/dia (DQ). Após 120 dias, o jejuno foi coletado e fixado na solução de Zamboni por 18 horas. Após a obtenção de cortes em criostato, a imuno-histoquímica foi realizada para marcar 5-HT e caspase-3. A quantificação de células imunorreativas (IR) à 5-HT e caspase-3 foram realizadas na lâmina própria, vilosidades e criptas. RESULTADOS: A condição diabética ocasionou um aumento do número de células 5-HT-IR nas vilosidades e criptas, enquanto que na lâmina própria houve uma redução dessas células, no jejuno de ratos STZ. Nos animais diabéticos, foi observada uma densidade aumentada de células apoptóticas no epitélio do jejuno, tanto nas vilosidades quanto nas criptas, por outro lado um número reduzido de células caspase-3-IR foi observado na lâmina própria. Possivelmente, a suplementação de quercetina suprimiu ligeiramente os fenômenos de apoptose no epitélio de vilosidades e criptas do jejuno de ratos STZ, no entanto, o efeito oposto foi observado nas células 5-HT-IR da lâmina própria. A suplementação com quercetina em animais saudáveis promoveu poucas alterações na função serotoninérgica e nos estímulos apoptóticos. CONCLUSÃO: Estes resultados sugerem que a suplementação de quercetina melhorou principalmente a função serotoninérgica afetada pelo diabetes, talvez devido às propriedades antioxidantes e anti-inflamatórias da quercetina.

ABSTRACT

To explore the mechanistic basis behind smooth muscle relaxant prospective of Bismarckia nobilis in gastrointestinal, respiratory and cardiovascular ailments. The methanolic extract of B. nobilis and sub-fractions have been evaluated in vitro rabbit isolated tissues, in vivo castor oil-induced diarrhea in rats and charcoal meal activity in mice. The B. nobilis extract relaxed spontaneous and K+(80 mM)- induced contractions in rabbit isolated jejunum preparations, CCh (1 µM) and K+ (80 mM)-induced contractions in tracheal and bladder preparations, PE (1 µM) and K+ (80 mM)-induced concentrations in aorta preparations, likewise verapamil. Spasmolytic activity of dichloromethane fraction is stronger as compared to aqueous fraction. In vivo castor oil-induced diarrhea in rats and charcoal meal activity in mice further supported spasmolytic activity. B. nobilis extract possess anti-spasmodic, anti-diarrheal, airway relaxant and vasodilator activities possible mediated through calcium channel blocking mechanism, justifying therapeutic utility of B. nobilis in diarrhea, asthma and hypertension.


El objetivo de trabajo fue explorar el mecanismo de acción relacionado con el efecto relajante del músculo liso inducido por Bismarckia nobilis (B. nobilis) en enfermedades gastrointestinales, respiratorias y cardiovasculares. El extracto metanólico de B. nobilis y subfracciones fue evaluado in vitro en tejidos aislados de conejos. Además se evaluó diarrea in vivo inducida con aceite de ricino en ratas y la actividad de harina de carbón vegetal en ratones. El extracto de B. nobilis relajó tanto las contracciones espontáneas como las inducidas por K+(80 mM) en preparaciones de yeyuno aisladas de conejos, las contracciones inducidas por PE (1 µM) y K+(80 mM) inducidas en preparaciones de aorta; de manera similar a verapamilo. La actividad espasmolítica de la fracción de diclorometano es más potente en comparación con la fracción acuosa. La diarrea inducida in vivo por el aceite de ricino en ratas y la actividad de la harina de carbón vegetal en ratones apoyaron aún más la actividad espasmolítica. El extracto de B. nobilis posee actividades antiespasmódicas, antidiarreicas, relajantes de las vías respiratorias y vasodilatadoras, posibles a través del mecanismo de bloqueo de los canales de calcio, lo que justifica la utilidad terapéutica de B. nobilis en la diarrea, el asma y la hipertensión.

ABSTRACT

Taurine (2-aminoethanesulfonic acid) is widely distributed in animal tissues and has diverse pharmacological effects. However, the role of taurine in modulating smooth muscle contractility is still controversial. We propose that taurine (5-80 mM) can exert bidirectional modulation on the contractility of isolated rat jejunal segments. Different low and high contractile states were induced in isolated jejunal segments of rats to observe the effects of taurine and the associated mechanisms. Taurine induced stimulatory effects on the contractility of isolated rat jejunal segments at 3 different low contractile states, and inhibitory effects at 3 different high contractile states. Bidirectional modulation was not observed in the presence of verapamil or tetrodotoxin, suggesting that taurine-induced bidirectional modulation is Ca2+ dependent and requires the presence of the enteric nervous system. The stimulatory effects of taurine on the contractility of isolated jejunal segments was blocked by atropine but not by diphenhydramine or by cimetidine, suggesting that muscarinic-linked activation was involved in the stimulatory effects when isolated jejunal segments were in a low contractile state. The inhibitory effects of taurine on the contractility of isolated jejunal segments were blocked by propranolol and L-NG-nitroarginine but not by phentolamine, suggesting that adrenergic β receptors and a nitric oxide relaxing mechanism were involved when isolated jejunal segments were in high contractile states. No bidirectional effects of taurine on myosin phosphorylation were observed. The contractile states of jejunal segments determine taurine-induced stimulatory or inhibitory effects, which are associated with muscarinic receptors and adrenergic β receptors, and a nitric oxide associated relaxing mechanism.

ABSTRACT

Objetivou-se identificar o perfil sociodemográfico de idosos vítimas de trauma, caracterizar doenças preexistentes e medicamentos utilizados no domicílio; calcular índices de trauma e desfecho clínico. Estudo retrospectivo e exploratório, com a análise de dados secundários de um banco de dados de um hospital geral terciário, entre 2008 e 2010. Foram estudados 131 idosos, média de idade 69,9 anos, 73,3% homens, 55,1% casados, 54,7% aposentados; 65,6% possuíam doenças preexistentes e 48,9% usavam medicamentos no domicílio. Houve representatividade de quedas (31,3%), seguidas por atropelamento (28,2%), com cabeça/pescoço sendo a região mais acometida (59,5%). Prevaleceu o trauma moderado (44,3%), com condições de sobrevida após o evento (80,2%). Houve associação entre mecanismo do trauma e doença preexistente (p=0,01) e entre mecanismo do trauma e sexo (p=0,03). O conhecimento das variáveis envolvidas com idosos vítimas de trauma possibilita aos profissionais de saúde o planejamento de medidas preventivas, visando aprimorar sua assistência.


The objective was to identify the sociodemographic profile of the elderly victims of trauma, to characterize preexisting conditions and medications taken at home, and to calculate indices of trauma and clinical outcomes. This is a retrospective and exploratory analysis from a database of a general hospital between 2008 and 2010. There were studied 131 elderly, mean age 69.9 years, 73.3% male, 55.1% married, 54.7% retired, 65.6% had preexisting conditions and 48.9% used drugs at home. There was a representative number of falls (31.3%), followed by running over (28.2%), with the head/neck region being the most affected (59.5%). Moderate trauma prevailed (44.3%), with conditions of survival after the event (80.2%). There was an association between mechanism of trauma and preexisting disease (p=0.01) and between mechanism of trauma and sex (p=0.03). The knowledge of the variables involved with the elderly victims of trauma enables healthcare professionals to plan preventive measures aimed at improving the assistance. Se objetivó identificar el perfil sociodemográfico de ancianos víctimas de trauma, caracterizar condiciones preexistentes y medicamentos tomados en casa, y calcular índices de trauma y evolución clínica. Se realizó un análisis retrospectivo y exploratorio de una base de datos de un hospital general terciario entre 2008 y 2010. Se estudiaron 131 ancianos, media of 69,9 años, 73,3% hombres, 55,1% casados, 54,7% jubilados, 65,6% tienen condiciones preexistentes y 48,9% estaban tomando medicación en casa. Hubo representación de las caídas (31,3%), seguido de atropello (28,2%). La región cabeza/cuello fue el más afectado (59,5%). Prevaleció trauma moderado (44,3%), con condiciones de supervivencia después del evento (80,2%). Se observó una asociación entre mecanismo de lo trauma y enfermedad previa (p=0,01) y entre mecanismo de lo trauma y sexo (p=0,03). El conocimiento de las variables que intervienen con ancianos víctimas de trauma permite a los profesionales de la salud planificar medidas preventivas para mejorar su asistencia.

With an aim to devise a prophylactic and/or therapeutic approach for preventing internalization of radiothallium (201Tl), and more importantly by implication, its chemical analogue radiocesium (137Cs) during any nuclear emergency, different ex vivo and in vivo animal models were created to determine the role of pH in absorption of 201Tl across jejunum/muscle tissue and whole body retention of 201Tl respectively. Movement of Tl+ under simulated pH conditions proved that pH had direct influence on its absorption. Oral intake of acidified water or parenteral administration of lactic acid was able to reduce the body burden of 201Tl by up to 12 and 50% respectively. The results indicate that acidification of gut, within physiological range may be used as an option for decorporation/inhibition of incorporation of radiothallium and radiocesium, particularly in cases of mass casualty.

OBJETIVO: Comparar a resistência cicatricial de anastomoses e de segmentos íntegros jejunais de ratos, submetidos à administração de vitamina C, em distintos períodos pós-operatórios. MÉTODOS: Foram estudados 50 ratos Wistar, submetidos a enterotomia seguida de anastomose término-terminal de segmento jejunal, a 10 cm da flexura duodenojejunal. Os animais foram distribuídos em dois grupos (n=25): Grupo I - controle; Grupo II - administração de vitamina C oral 100 mg/kg. Avaliaram-se as pressões de ruptura anastomótica e do segmento íntegro jejunal nos 3º, 5º, 7º, 21º E 28º dias do pós-peratório. RESULTADOS: Os ratos que receberam vitamina C apresentaram pressão de ruptura anastomótica maior nos 5º, 7º, e 28º dias pós-operatórios. O mesmo ocorreu com as pressões de ruptura do segmento íntegro jejunal dos ratos. CONCLUSÃO: A vitamina C aumentou a resistência das anastomoses jejunais dos ratos, tanto no pós-operatório imediato quanto no tardio. Além disso, a resistência final dos segmentos jejunais íntegros dos ratos submetidos à administração de vitamina C foi significativamente maior do que no Grupo Controle.


OBJECTIVE: To compare the resistance of anastomosed and intact jejunal segments of rats submitted to administration of vitamin C in different postoperative periods. METHODS: Fifty Wistar rats underwent enterotomy followed by end-to-end anastomosis of the jejunal segment, 10 cm from the duodenojejunal flexure. The animals were divided into two groups (n = 25): Group I - control; Group II - administration of oral vitamin C 100 mg/kg. We evaluated the bursting pressures of the anastomotic and the intact jejunal segments in the third, fifth, seventh, 21st and 28th postoperative days. RESULTS: The rats that received vitamin C had higher anastomotic bursting pressure in the fifth, seventh and 28th postoperative days. The same happened with the bursting pressures of intact jejunal segments. CONCLUSION: Vitamin C increased the resistance of jejunal anastomoses in rats, both in the immediate and in late postoperative periods. In addition, the final resistance of intact jejunal segments of rats under administration of vitamin C was significantly higher than in the control group.

OBJETIVO: Comparar a resistência cicatricial de anastomoses jejunais em ratos, submetidos à administração de vitamina C e de hidrocortisona, em distintos períodos pós-operatórios. MÉTODOS: Foram estudados 40 ratos Wistar, submetidos à secção e subsequente anastomose término-terminal de segmento jejunal, a 10 cm da flexura duodenojejunal. Os animais foram distribuídos em quatro grupos (n=10): Grupo I - controle; Grupo II - administração de vitamina C oral 100 mg/kg; Grupo III - administração de hidrocortisona intraperitoneal 10 mg/kg; Grupo IV - administração de vitamina C mais hidrocortisona nas doses e vias de administração acima. Avaliaram-se as pressões de ruptura anastomótica no 5º e 21º dias do pós-operatório. RESULTADOS: Os ratos que receberam vitamina C isolada ou associada a hidrocortisona tenderam a ter pressão de ruptura maior do que os demais grupos, tanto no 5º quanto no 21º dia pós-operatório. CONCLUSÃO: A vitamina C contribui para aumentar a resistência das anastomoses jejunais dos ratos durante os primeiros cinco dias do pós-operatório. A resistência das anastomoses jejunais murinas foi pouco influenciada pela administração de corticóide intraperitoneal.


OBJECTIVE: The effects of vitamin C and hidrocortisone on anastomotic healing process are controversial. The objective of the present paper was to compare the jejunal anastomotic tension in different postoperative periods. METHODS: 40 male rats weighing 200 to 400 grams were submitted to laparotomy. The jejunum was transversally cut 10 cm from the duodenojejunal flexure, and subsequently anastomosed. The rats were divided into four groups (n=10). Group I - control, Group II - oral administration of vitamin C (100 mg/kg), Group III - intraperitoneal administration of hidrocortisone (10 mg/kg), and Group IV - administration of vitamin C and hidrocortisone at the above mentioned doses. The anastomotic resistance was determined by using bursting pressure test on the 5th and 21st postoperative days. RESULTS: The resistances of the groups with vitamin C, isolated or associated to hidrocortisone trend to be higher in both postoperative periods. However, the administration of intraperitoneal steroid did not change the resistance of murine jejunal anastomosis. CONCLUSION: The results of the present study suggest that vitamin C enhances the resistance of jejunal anastomosis in the rat.

ABSTRACT

This study was carried out to evaluate the effects of decoction of Syrian cuminum seeds [1:3 W/V] was evaluated on the isolated rabbit's jejunum. 2 ml of the extract was added to the [100mL] nutrient solution of the jejunum. Two parameters were used to evaluate the effect of cuminum extract: the frequency and width of the intestinal motility within 5 mints on the strip. Cuminum extract showed a significant inhibition on the width of the motility of isolated rabbit's jejunum [P< 0.05]. However this extract had no significant effect of the frequency of the motility. Cuminum cyminum water extract can inhibit the motility of rabbit's jejunum that support the traditional use of this plant as antispasmodic herb in the Syrian traditional medicine

ABSTRACT

The non-steroidal anti-inflammatory drug [NSAID] indomethacin causes, via its adverse effects, damage to the gastrointestinal mucosa of humans and experimental animals. The indomethacin-induced intestinal injury in mice jejunum is considered an experimental model of Crohn's disease, as one of the inflammatory bowel diseases [IBD's]. The semi-essential amino acid arginine, which is a precursor of nitric oxide, is proposed to promote gastrointestinal mucosal integrity. The present work aimed to study the possible protective role of dietary supplementation with arginine in ameliorating indomethacin-induced mucosal injury of mice jejunum. The present study was carried out on forty adult male mice which were divided into 4 equal groups; group I [negative control group] which received no treatment, group II [positive control]; mice received dietary L-arginine alone in a daily dose of 300 mg/Kg, group III [indomethacin group]; mice of this group received 2 consecutive subcutaneous injections of indomethacin in a dose of 7.5 mg/kg, 24 hours apart and group IV [arginine group]; in which arginine supplementation was provided 2 days before the administration of indomethacin, maintained during the administration and continued 3 days later till the end of the experiment. Mice of all groups were sacrificed by the end of the 7[th] day and specimens from the jejunum were dissected and processed for light and scanning electron microscopic examinations. Indomethacin-treated group exhibited jejunal mucosal injury. Light microscopic examination of this group, using H and E and toluidine blue- stained semithin sections showed distorted villi and sloughing of some of their apical parts with extrusion of many degenerated cells, intermingled with excess mucous. Some enterocytes appeared degenerated with loss of their regular arrangement, while the goblet cells appeared distended with excess mucous secretion. Increased cellular infiltration and edema of the villous core and in the lamina propria between the glands were noticed. Increased mucous secretion was also demonstrated by combined alcian blue-periodic acid Schiff reaction. Scanning electron microscope revealed alteration in the architecture of many villi which appeared short, blunt or with denuded surface and occasionally covered with membrane-like structure. The group of mice received arginine [group IV] revealed restoration of mucosal integrity in the form of regular villi with intact epithelial coverings including enterocytes and goblet cells. Some villi appeared shorter, while others showed partially denuded apical surface. Arginine evoked a remarkable cellular infiltration. Lymphocytes and macrophages were among the infiltrating cells and their roles were suggested to be vital in the healing process. Dietary L-arginine provided satisfactory protection against indomethacin-induced mucosal injury in mice, most probably via its role as a nitric oxide donor. So supplementation with dietary arginine is recommended

ABSTRACT

OBJETIVO: Estudar a taxa de proliferação celular no jejuno e nas células epiteliais das criptas do intestino grosso em ratos pinealectomizados imediatamente após o nascimento. MÉTODOS: 24 ratos machos Wistar foram divididos em dois grupos. Grupo agudo (n=12) e Grupo Crônico (n=12). Seis animais de cada grupo foram operados para remover-se a glândula pineal (Pinealectomia-PnX), e outros seis animais foram controle (sham pinealectomia-C). Os animais de ambos os grupos foram sacrificados 15 e 90 dias após a cirurgia, respectivamente. RESULTADOS: No grupo agudo, a pinealectomia dos ratos não causou alterações significativas na proliferação celular (PnX=58,77±1,77 e C=60,88±1,10 no cólon descendente / PnX=31,56±0,45 e C=31,73±0,47 no jejuno proximal) e na população celular de criptas (PnX=24,92±4,82 e C=23,60±2,48 no cólon descendente / PnX=39,92±3,49 e C=44,32±5,56 no jejuno proximal). Contudo, no grupo crônico houve aumento na proliferação celular das criptas no jejuno proximal (PnX=57,54±2,19 e C=47,19±7,3), e no cólon descendente (PnX=37,78±2,22 e C=17,92±2,28). CONCLUSAO: Como o aumento epitelial celular das criptas intestinais no grupo crônico pode ser avaliado como fator predeterminante da carcinogênese, faz-se necessário o conhecimento da interação entre esta glândula e este evento.

ABSTRACT

OBJETIVO: Investigou-se o efeito de ácidos graxos de cadeia curta (SCFA) na mucosa intestinal na presença de lesão por isquemia e reperfusão (IRI). MÉTODOS: Foram criados seis sacos fechados no intestino delgado (três no jejuno e três no íleo) em 10 ratos Wistar. Ao sacos laterais de ambas as regiões intestinais foram submetidos a IRI (15/15 minutos) enquanto que o saco medial não sofreu interrupção do suprimento sanguíneo. Nos sacos laterais ambas as regiões injetou-se SCFA ou solução fisiológica na luz intestinal. Nos sacos mediais não se injetou nenhuma solução. RESULTADOS: Tanto no jejuno quanto no íleo o escore de injuria da mucosa intestinal foi mais alto nos sacos tratados com solução salina do que nos controles. Os sacos que receberam SCFA apresentaram menor escore inflamatório no íleo (p=0.03) porém sem diferença no jejuno (p=0.083) quando comparados com os sacos injetados com solução salina. Observou-se um significante maior acumulo de neutrófilos nos sacos tratados com solução salina (p < 0.01) do que nos outros dois sacos em ambas as regiões. CONCLUSAO: Os SCFA protegem a mucosa intestinal distal e diminuem o acumulo de neutrófilos na lamina própria após IRI.

ABSTRACT

OBJETIVO: Investigar os efeitos do uso combinado da glutamina (GL) e do hormônio do crescimento (GH) no intestino de ratos submetidos a ressecção de 80% do intestino delgado. MÉTODOS: Vinte e quatro ratos Wistar foram randomizados para receber uma a dieta padrão- grupo controle (GC, n=12) ou a mesma dieta adicionada de glutamina 4% (isocalórica, isoproteica) - grupo glutamina- hormônio do crescimento (GL-GH, n=12) após a enterectomia à 80%. Este último grupo recebeu por via sub-cutânea, 0,6 UI/dia de GH. Grupos de seis ratos cada foram sacrificados no 5º e 14º dias. As seguintes variáveis foram estudadas: peso corporal, peso de mucosa, histomorfometria e conteúdo de DNA no segmento ressecado inicialmente e no intestino adaptado coletado após o sacrifício. RESULTADOS: Todos os animais perderam peso até o 5º dia, estabilizando-se após esta data em ambos os grupos. Não houve diferença estatística no peso da mucosa associada a grupos ou datas. O peso da mucosa do íleo diminuiu dos dados iniciais para os finais, quando comparados a mucosa jejunal (p<0.02). O conteúdo de DNA aumentou dos dados iniciais para os finais (p=0.001) em ambos os grupos, porém, o aumento foi maior nos animais do grupo GL-GH (CG = 0.53 [95% CI, 0.44-0.62] g/cm-1 vs. GL-GH= 0.85 [95%CI, 0.76-0.94] g/cm-1; p<0.01), especialmente no 14º dia. O conteúdo de DNA no íleo foi significativamente maior que no jejuno (p=0.01). Houve um aumento significativo na espessura da parede e na profundidade da cripta, no grupo controle (p<0.01). CONCLUSÃO: A adaptação intestinal após ressecção extensa é melhorada com o uso combinado de glutamina e GH.

ABSTRACT

Effects of the aqueous extract of T. sessilifolius on the gastrointestinal muscle were investigated on smooth muscle preparations isolated from rabbit jejunum, guinea pig ileum and on gastrointestinal transit in mice. Elemental analysis of the extract was also carried out. The aqueous extract of T. sessilifolius evoked a concentration dependent contraction of the rabbit jejunum and guinea pig ileum. The contractions evoked by the extract were not attenuated either by atropine or mepyramine, but they were completely blocked by verapamil. The elemental analysis revealed the presence of Mg, Zn, Fe, Cu, and very high concentration of Ca. The intraperitoneal LD50 in mice was found to be 1500 mg/kg. The aqueous extract of T. sessilifoliius possesses active components that may be mediating the observed biological activity through calcium mobilization.

BACKGROUND: The effects of vitamin C on anastomotic healing process are controversial. OBJECTIVE: To compare the jejunal anastomotic tension and in the upright segment in different postoperative periods. METHOD: Fifty male rats weighing 250 to 400 grams were submitted to laparotomy. The jejunum was transversally cut 10 cm from the duodenojejunal flexure, and subsequently anastomosed. The rats were divided into two groups (n = 25). Group I--control, Group II--oral administration of vitamin C (100 mg/kg). The anastomotic and the upright segment resistance was determined by using bursting pressure test on the 3rd, 5th, 7th, 21st and 28th postoperative days. RESULTS: The rats submitted to oral administration of ascorbic acid show higher bursting pressure on the 5th, 7th and 28th postoperative days. The bursting resistances of the upright segment was higher on the rats submitted to vitamin C ingestion. CONCLUSIONS: Vitamin C enhances the anastomotic and jejunal resistances. Moreover, the final resistance on the upright jejunal segment was significant higher than in the control group.

ABSTRACT

Intestinal protection in mice against radiation injury by WR-2721 (300 mg/kg body wt, i.p., 30 min before irradiation) was studied after whole body gamma irradiation (0.5, 1.5, 3.0, 4.5, or 6.0 Gy). Crypt survival and induction of apoptosis, and abnormal mitoses in crypt cells in the jejunum were studied on day 1, 3 and 7 after irradiation. Irradiation produced a significant decrease in crypt survival, whereas apoptosis and abnormal mitoses showed a significant increase from sham-treated control animals. Maximum changes in all the parameters were observed on day 1 after irradiation and the effect increased linearly with radiation dose. There was recovery at later intervals, which was inversely related to radiation dose. WR-2721 pre-treatment resulted in a significant increase in the number of surviving crypts, whereas the number of apoptotic cells in the crypts showed a significant decrease from respective irradiated controls on day 1 after exposure. The recovery was also faster in WR-2721 pre- treated animals. It is concluded that WR-2721 protects against gastrointestinal death by reducing radiation induced cell death, thereby maintaining a higher number of stem cells in the proliferating compartment.

Glutamine is the most abundant amino acid in the blood and plays a key role in the response of the small intestine to systemic injuries. Mucosal atrophy is an important phenomenon that occurs in some types of clinical injury, such as states of severe undernutrition. Glutamine has been shown to exert powerful trophic effects on the gastrointestinal mucosa after small bowel resection or transplant, radiation injury, surgical trauma, ischemic injury and administration of cytotoxic drugs. Since no study has been performed on the malnourished animal, we examined whether glutamine exerts a trophic effect on the intestinal mucosa of the malnourished growing rat. Thirty-five growing female rats (aged 21 days) were divided into 4 groups: control - chow diet; malnutrition diet; malnutrition+chow diet; and malnutrition+glutamine-enriched chow diet (2 percent). For the first 15 days of the experiment, animals in the test groups received a malnutrition diet, which was a lactose-enriched diet designed to induce diarrhea and malnutrition. For the next 15 days, these animals received either the lactose-enriched diet, a regular chow diet or a glutamine-enriched chow diet. After 30 days, the animals were weighed, sacrificed, and a section of the jejunum was taken and prepared for histological examination. All the animals had similar weights on day 1 of experiment, and feeding with the lactose-enriched diet promoted a significant decrease in body weight in comparison to the control group. Feeding with both experimental chow-based diets promoted significant body weight gains, although the glutamine-enriched diet was more effective. RESULTS: The morphological and morphometric analyses demonstrated that small intestinal villous height was significantly decreased in the malnourished group, and this change was partially corrected by the two types of chow-based diet. Crypt depth was significantly increased by malnutrition, and this parameter was partially corrected by the two types of chow-based diet. The glutamine-enriched diet resulted in the greatest reduction of crypt depth, and this reduction was also statistically significant when compared with control animals. CONCLUSIONS: Enteral glutamine has some positive effects on body weight gain and trophism of the jejunal mucosa in the malnourished growing rat

ABSTRACT

This work aimed to evaluate the prospective effect of some antioxidants against MTX toxicity. 90 adult albino rats, kept on a standard diet and water, were divided into 5 groups. Group I served as a control group, group II received methotrexate [MTX] in a dose of 2.5 mg/kg b.wt. for 3 consecutive days, group III received MTX concomitantly with vitamin E 600 mg/kg, group IV received MTX concomitantly with vitamin A 5000 IU/kg, and group V received MTX concomitantly with both vitamin E and vitamin A. The results were statistically analyzed and revealed that jejunum of rats with MTX [group I] showed significantly short, wide villi and deep crypts, marked lymphocytic infiltration in epithelium and lamina propria of the villi. Union of the villi either at the bases or their tips was a protective mechanism to decrease the oozing of plasma from the tissue to the lumen. Goblet cells showed decreased intensity of alcian blue stain. Alkaline phosphatase reaction was reduced at the brush border of the villi despite of its increased serum level. The electron microscopic results revealed markedly swollen mitochondria, reduction in the number and size of microvilli at the brush border, the nuclei showed signs of degeneration. Laboratory investigation revealed a significant reduction of TLC, PC, a significant elevation of serum AST, ALT, ALP, urea and creatinine in group II when compared with control group. In the same time, there was significant improvement of the previous parameters in groups III, IV and V when compared with MTX group. Concomitant administration of vitamins E and A with MTX gave the best protection against MTX-induced damage of both intestinal mucosa and laboratory parameters

Os efeitos do cloreto de benzalcônio (CB) sobre o número de neurônios, a espessura da musculatura própria e o perímetro intestinal externo foram determinados após desenervaçäo aguda (até 10 dias pós-aplicaçäo do CB) e crônica (30 e 60 dias pós-aplicaçäo do CB) do jejuno proximal de ratos. Houve reduçäo significativa no número de neurônios mioentéricos em todos os segmentos tratados. A extensäo da desnervaçäo variou em funçäo do tempo decorrido após o tratamento: nos segmentos desnervados cronicamente o número de neurônios era superior àquele nos segmentos desnervados agudamente. Este achado pode ser resultado de fenômenos de neuroplasticidade ocorrendo durante a fase crônica. A desenervaçäo mioentérica induziu aumento da espessura da musculatura própria, especialmente da camada muscular longitudinal, sugerindo maior sensibilidade desta camada à desnervaçäo mioentérica

ABSTRACT

Chilli (containing the active ingredient capsaicin) forms an important flavouring agent in the preparation of meals in the tropics. Previous studies have shown that capsaicin in high doses causes gross structural and functional changes in the gut. The present study investigates the effect of pure capsaicin on the absorption of glucose, water and alanine by the small intestine of the rat. Perfusion studies were carried out using a 10 cm jejunal segment. Absorption rates of glucose, water and alanine from a control solution containing the nutrient and from a test solution containing added capsaicin were compared. Recovery of absorptive function of the intestinal mucosa after exposure to capsaicin was also studied. Absorption of water, glucose and alanine was found to be significantly reduced in the presence of capsaicin. Recovery of absorptive function occurred when capsaicin was withdrawn from the perfusate. It was concluded that capsaicin adversely affected absorption of nutrients from the rat small intestine; this effect was reversible at least in the case of some nutrients.

Experiments were carried out to determine the advantage in using isolated rat jejunum in the assay of acetylcholine (ACh) in preference to other tissues. Rat jejunum was found to be sensitive to ACh 0.01 micrograms, 5-hydroxytryptamine (5HT) 0.2 to 0.5 microgram and least sensitive to histamine. (Ach greater than 5HT greater than histamine).
